Overview

Isolate highly purified human CD19+ cells from whole blood or buffy coat and leukoreduction system chamber (LRSC; also known as an LRS cone) samples by immunomagnetic positive selection, with the EasySep? HLA Chimerism Whole Blood CD19 Positive Selection Kit. Widely used in published research for more than 20 years, EasySep? combines the specificity of monoclonal antibodies with the simplicity of a column-free magnetic system.

In this EasySep? positive selection procedure, desired cells are labeled with antibody complexes recognizing CD19 and magnetic particles. The cocktail in this kit also contains an antibody to human Fc receptor to prevent non-specific binding. Labeled cells are separated using an EasySep? magnet and by simply pouring or pipetting off the unwanted cells. The cells of interest remain in the tube. Following magnetic cell isolation, the desired CD19+ cells are ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. The CD19 antigen is present on mature B cells and B cell progenitors but is lost upon maturation to plasma cells. It is also expressed on follicular dendritic cells.

Starting with human whole blood, the CD19+ cell content of the isolated fraction typically ranges from 94.3 - 99.6% (as assessed by staining the start and isolated fractions with anti-CD19 or anti-CD20 antibodies, respectively). In the above example, the purities of the start and the final isolated fractions are 5.0% and 98.6%, respectively (gated on CD45).
NOTE: Red blood cells were removed from the start sample by lysis prior to flow cytometry.
